The tech giant's move signals its intention to strengthen its foothold in the AI sector.
This latest investment follows Amazon's initial pledge of $1.25 billion in September, bringing its total commitment to Anthropic up to $4 billion.
Amazon will maintain a minority stake in Anthropic and refrain from securing a board seat, in line with the terms established during the AI startup's last valuation at $18.4 billion.
Anthropic, founded by former OpenAI research executives and employees, has raised a total of approximately $7.3 billion across five funding rounds.
